Nepal and Guinea established formal diplomatic relations on May 12, 2016. The Permanent Representative of Nepal to the United Nations (UN), Durga Prasad Bhattarai, and Permanent Representative of the Republic of Guinea to the United Nations (UN), Mamadi Touré signed the agreement at the Permanent Mission of Nepal in New York.

The dignitaries also informed Ban Ki-Moon, Secretary General of UN, about the establishment of relations. Multilaterally, Nepal and the Republic of Guinea are part of G-77, and the Least Developed Countries. The relations between the countries have been friendly and cordial.
